SYNTHESIS DESIGN OF A NATURALLY OCCURRING ANTI-FUNGAL MACROLIDE ANTIBIOTIC (-)-PYRENO-PHORIN: A RETROSYNTHETIC APPROACH

Abstract

Synthesis is the fundamental enabling discipline of natural product chemistry without it the development of rarely found essential natural products cannot progress from their characterization to bio-information and ultimately for pharmaceutical implementation. Designing synthetic routes to these natural products through the strategic application of retrosynthetic analysis/synthon disconnection approach, pioneered by prof.E.J.Corey of Harvard University has emerged as powerful tool in modern natural product chemistry. Keeping a bird’s eye view on the works published in journals and patent literatures, some synthesis schemes have been proposed for a naturally occurring anti-fungal antibiotic (-)-Pyrenophorin in a novel way basing on the retrosynthetic analysis. The proposed synthesis planning being a theoretical exploration, the actual laboratory execution requires the cross examination of a considerable number of factors such as reactions, reagents and order of events. In actual practice, generally that route is most feasible which satisfies the specific criterion for an ideal synthesis.
